Belantamab mafodotin in real-world practice: ocular care matters

In the DREAMM-2 trial, belantamab mafodotin (belamaf) monotherapy showed clinical activity and a manageable safety profile in pretreated patients with relapsed/refractory multiple myeloma (RRMM). Real-world evidence on this population is scarce. A new study in Annals of Hematology, co-authored by Antje Mevius (IPAM e.V.), describes patients treated with belamaf in Germany between August 2020 and March 2023, based on anonymised health insurance claims data.

Key findings

• 31 heavily pretreated patients with RRMM were included (median age 66 years).

• Ophthalmological visits increased across the study periods: 32.3% before, 51.6% during and 68.2% after belamaf treatment.

• During belamaf treatment, 22.6% of patients had one or more new ocular diagnoses of interest, most frequently corneal-related events; keratitis was the most frequent ocular diagnosis overall.

• Real-world progression-free survival was 4.3 months and overall survival 12.3 months.

The data emphasise the importance of ocular supportive care during belamaf treatment in routine practice and show that RRMM remains an aggressive disease with a poor prognosis.
